Pro Tools is, hands down, the music industry standard. However, the file has to be in a certain file format (midi, etc) in order to split out the components - such as vocals and beats.

If its an audio file such as a wav or mp3 you can use something like sound forge which has filters etc.
You'd have to do it in 2 goes, first open the file and filter out everything except the beats and save that. Then open the original file again and filter out everything except the vocals and save that.

You will never be able to completely seperate the sections however - it's a matter of taking out the bass of the sample in order to have as much of the vocal on its own, and taking out most of the highs for drums. Because of this the quality of the vocal/drums will not be great.
