Okay, very simple.
Offense - wants to score.
Defense - wants to stop other team from scoring.
Offense has 4 chances to move the ball 10 yards. Then they get more chances. If they fail, the defense usually gets the ball, and that teams offense takes the field and the cycle starts over again.
When one team crosses the goal line, after passing and handing the ball off to their friends, they get a touchdown.
Simple explanation!

11 players from each team take the field for a total of 22 players on the field at a time. One team is on the "offense", while the other is the "defense".
The defense tries to keep the offense from scoring.
The offensive team's goal is to score. The offense scores when it
1) gets the ball into the endzone - aka Touchdown - worth 6 points.
2) kicks the ball through the uprights. If the ball is kicked through the uprights immediately after a touchdown, it is known as an extra point and worth 1 point. If the ball is kicked through the uprights not following a touchdown, it is known as a field goal (3 points)
So how does the offense move the ball from one end of the field to the other?
When the offense gets the ball, they have four opportunities to move the ball ten yards towards the endzone. These opportunities are known as "downs". There are four downs - 1st down, second down, third down and fourth down. If the team cannot move the designated ten yards in the four tries, then the other team takes over the ball and the team that was on offense goes on defense, and the team on defense goes on offense.
For example, Team O is on offense. Let's say Team O is starting at the fifty yard line. Since the field is 100 yards long, Team O has fifty yards to go until it reaches the endzone. Team O has a "first and ten" on the fifty yard line. This means that the team has four tries to get to the forty yard line. (50 - 10 = 40) They can move the ball by running it, or by passing it.
1st and 10, fifty yard line - Team O hands the ball to the running back, who runs towards the end zone. However, Team D is there to meet him, and Team O's runnning back only is able to run with the ball three yards closer to the end zone. So now, the ball is at the forty seven yard line.
2nd and 7, forty seven yard line. It is second down, because this is the teams second chance to get to the forty yard line. The number 7 means that there are seven more yards to the forty yard line. It's 2nd and 7 and Team O decides it wants to throw the ball. The QB drops back to pass and throws the ball to his "wide receiver". However, the wide receiver cannot hold onto the ball and it falls to the ground. This is known as an incomplete pass. Team O has not gotten any closer to the 40 yard line, and has used another chance. This means that it is 3rd and 7. (instead of 2nd and 7)
3rd and 7, still on the 47 yard line. Team O decides to pass again. The QB drops back and throws the ball to his "tight end". The tight end for Team O catches the ball all the way down at the 20 yard line where he is tackled by the defense. Team O got the ball well beyond the 40 yard line that Team O had to get to. What does this mean? This is known as a "first down". This means that because Team O was able to get the ten yards it needed, or in this case a lot more, it gets "a fresh set of downs" and four more chances to go the next ten yards. Because the tight end was tackled at the 20 yard line, that is where the ball is placed. So now, Team O has "1st and 10" at the 20 yard line. This means that Team O has four chances to get the ball to the ten yard line (20 - 10 = 10).
Let's say that Team O throws the ball on the 1st and 2nd downs. In both cases, the Wide receiver for Team O can't make the catch. This means that it is 3rd and 10 for Team O. Team O decides to throw again on 3rd down, but as the QB for Team O drops back to throw the ball, a member of Team D tackles the QB before he can throw the ball or run it forward at all. This is known as a "sack". When the defense sacks the QB, the ball is placed wherever the QB is tackled. A lot of times, this means that instead of gaining yards, the QB loses yards. For Team O, the QB was sacked at the 25 yard line. It is now 4th down and Team O still needs to get to the 10 yard line, meaning that it is "4th and 15 (25 - 10 = 15)." Usually, a team will either punt the ball (meaning kick it to the other team) or kick a field goal (3pts). In this case, Team O is close enough to the end zone that they try to kick a field goal. Team O kicks the ball through the uprights and is given 3 points.
Now that Team O has scored, Team O will kick to Team D. Team D will now get a chance to go on offense and Team O will play defense.
I know it seems like a lot, but my suggestion would be to try and watch a football game on TV. John Madden is particulary good to listen to during a game, because he explains a lot of the more obvious aspects of the game. This will help you as a beginner.

If not, talk to a girl friend who likes football and can explain some things toyou..
Either that, or ask a guy to help you learn about it. Lots of guys wouldn't mind explaining football to a cheerleader!
