Can I isolate the bacteria in a petri dish or a test tube? or is there some other method?

I am in microbiology and im doing an experiment.. so its not playing around

take a swap dip it into sterile water
rub the swab up and down on your hand (the safest way)
cover a strip about an inch wide and two inches long
place the swab in an tube of liquid broth incubate for two days
use an innoculating loop and innoculate the plate (get your instructor to show you the proper method (always use the proper safety equipment). If the broth is really cloudy use a 1/1000th disposable loop. incubate the plate 2 days. You should be able to pick off indivual pure colonies in the 3rd or 4th quadrant. These colonies can be tranfered to another plate, or placed into a broth tube. Hope this helps. Pardon mispellings its late and spell check is not working

It is really easy to grow bacteria. Just take some milk, put in a spoonful of yogurt and let it sit a day or two. Bingo! lots of bacteria. Or if you prefer you could just use soup.

You realize that this amount of bacteria would make you sick, right? So, it is not a good idea to play around with bacteria. That;s why we have refrigerators, sanitary canning and bottling plants, and soap and water to wash our hands and clothes and dishes.

Bacteria are pretty cool, but if you want to learn about how to handle them safely, go to college and take a course in microbiology.

You might try putting them in a tube filled with Luria broth and putting the tube in a shaker at 37 deg C overnight. That's if you just want a lot of them.

Use a petri dish to start your culture, use a nutrient medium like PDA or V8. Separate your colonies to separate dishes after a few days.
