Port 25 is used as your outgoing mail port. The blocking takes place – 99% of the time at least – on your ISP’s side. ISPs do this to ensure they can control the flow of mail coming out of their systems; monitor excessive mails, potential viruses, commercial spammers, etc.

To see if it is anything on your system you can stop any firewalls and try again. Has anything on your connection or your service changed over the past 3 weeks?

I would seriously consider calling your provider and verifying any issues on their end or with your account. The ISP I worked for would often block port 25 (outgoing mail) on any system it discovered was infected with a Trojan and/or sending out massive amounts of unsolicited e-mail.
