If you mean to shrug the shoulders, like to lift them up a bit, sometimes accompanied by the head pulling down a bit , there is no rule, many gestures in western culture are less of a rule than anything. But when it's done, you are correct, it signifies anything from mild uncertainty to outright "i don't know". but if you DON'T do it while you are using words that mean "i'm not sure" you will still be understood, and no one will wonder why you didn't also shrug.

The word is shrug...not shrink. I do not know the origin, but the definition of shrug is to raise (the shoulders) as an expression of dislike, doubt, indifference, or impatience.
