it's marcellus's soul. those kids stole it for the devil and he wants it back (the first time you see him is from behind and he has a plaster on the back of his head where it was taken out by the devil). this is also why the bullets miss samuel l jackson and john travolta: samuel is right - god really did intervene. also, travolta does not pull the trigger but the guy in the car has his head blown off. a vengeful god is intervening again.
when you know this and watch the film again, stuff makes more sense.

Diamonds!
I believe is the suitcase from the earlier movie Reservoir Dogs, the suitcase with the diamonds, this even if Tarantino has admitted that there is no official explanation behind the briefcase's contents, and that it was simply written into the screen play as an intriguing McGuffin.

I think the idea is that it is not supposed to be anything in particular so a myth will build around it and it will be more interesting for the audience. The same idea was used before in a movie called Repo Man by Alex Cox who did it better then tarantino who just took the idea straight from there but I'm sure it has been used in other movies too just can't think of them right now!

most people think that its wallaces soul (hence the plaster on the back of his neck, where is soul was sucked out from, and the combination 666 on the suitcase) suggesting that wallace was the devil. but tarantino is always interviewed about this, and hes said that its nothing, that us guys can think what we want about whats inside... who knows?!

I also have always thought it was Wallace's soul. The band aid on the neck is where the thing was sucked out of. I believe they stole it back from the devil and that is why all those horrible things happen around it. Now, what is Mullholland Drive even about?

the soul of marcellus wallace. Fell out of his head, thats why he has the plaster on the back of his neck. Heard this once and quite liked it. Don't know where it came from though.
