It's actually because of political cartoons that started around 1874 onto which one of the first was a donkey trying to throw a elephant down a hole . People started to adopt these symbols when others made cartoons using the same symbols with a sometimes different political meaning involved .

Other than the real, legitimate reason, I believe it has to do with the stubborn, plodding on, determination of the democratic party vs. the bungling, clumsy, bulky, (high cost of government) of the republican party. Donkey's are viewed as a dumb animal, yet, not so dumb when they don't want to be pushed past their limit. And, they work very smart. They are actually smarter than we give them credit for.
