I went to Jaipur from Delhi by scorpio. The distance by road is around 270 kms and it took me around 3-4 hours at 80-kms per hour. Jaisalmer is straight ahead and around 515 kms from Jaipur which will take you almost half a day or 12 hours by road. From Jaisalmer back to Delhi, you will have to come back by the same route which is 515kms by road till Jaipur and 270kms from Jaipur back to Delhi. So the total distance from Jaisalmer to Delhi is around (270kms + 515kms) 793 kms which is around 24hours or a full day's travel. You will have to go on a total of 3 National Highways (NH8, NH11 & NH15). No alternate route exists. For an exact idea of the road route, click on the link below.

There are many tourist spots along the way, like Fatehpur Sikri, Rajgarh and Bikaner. Dont miss Fatehpur Sikri, Lovely gardens and beautiful landscape. I went there in december last year after I visited Jaipur. Enjoy the trip

Jaisalmer has an airport (code JSA), but unfortunately there's no scheduled service at this airport.
According to the maps I have, there should be train service arriving at Jaisalmer, so you can check with the train service when you arrive there.
One way to cut time is by flying to a closer city. Your best choice would be Jodhpur (code JDH). Indian Airlines (IC) provides a daily flight from Jaipur (JAI) to JDH, and a daily flight from JDH to Delhi (DEL). From Jodhpur try taking a car or train to Jaisalmer and back again.
