This would be an easier question to answer if you would have given us a time frame.
The wheel? Printing press? Penicillin? Automobiles? Light bulb? Space travel? The computer? Microwave popcorn?
Pick one. They've all sginificantly changed the course of mankind. (Except maybe the popcorn, I'm just hungry. LOL)

Historically speaking, i believe the invention of the wheel has had the greatest impact on human civilization. It allowed our ancestors to travel farther and faster than they could have done by themselves or with animals, and it is hard to imagine us progressing much beyond the Stone Age without it.
(Note: fire was not invented. It just happens. Humans learned to *use* it.....but no human *invented* it.)

They always say the absolute most revolutionary one was the printing press. Without it, there would have been no successful protestant reformation in Europe and other important historical milestones in history.
