I use Ad-Aware, it's very good but doesn't catch everything. I suggest you also run Spybot Search & Destroy (it's also free) Between the two of them, you have solid anti-spyware defenses. Whatever one misses, the other one picks up.

Of course you need anti-virus software, too. I assume you already have one, but if not AVG Free edition works well, and is free. Hey, there's a trend here... :)

The best antispyware program I know of is webroot spyweeper- it even cleans rootkits, but costs a little bit (not quite free, $29.95 last I checked)

Using MS Windoz is asking for all sorts of trouble. Windoz is a big target of mal adjusted crazys wanting to hurt it and you. You really have to do a lot of work to keep Windoz clean of malware and running properly.

1. Ad-aware is said to be a very good program for cleaning spy and ad ware. Keep it up to date and it will find most ad programs. There has been some news that these types of programs have been under attack by lawyers for ad companies to allow their ads.
2. You also need a good anti-virus program. There are several good free programs. Yahoo/Google search will find them.
3. Windoz is always dirty. It picks up all sorts of trash. Search for and download Ccleaner. It is a very good cleaning program and it is free.
4. Defrag you hard drive regularly, especially after cleaning.

I use a Linux operating system and while it is rarely attacked by malware, it can import, store, and export malware. I have an anti-virus program so that I don't pass bad stuff on to Windoz users.

In a word, NO.

Pick and choose at least 2 from the list below. New spywares are created literally hourly, so no single anti-spyware can really detect all malware. Have at least 2 on hand and alternate scanning with each every few days or so. All are free. All can remove as well as detect.

Spybot Search and Destroy:

Ewido:

Adaware SE:

A-Squared:

Windows Defender (Beta 2):

Unfortunately, many "paid for" anti-spyware products are in fact either of limited usefulness or downright scams. When in doubt, look at the list of rogue anti-spyware solutions listed here:

And to protect your computer from ever getting infected with spyware in the first place, you can use these 2 freebies:

SpywareBlaster:

SiteAdvisor:

Of course you also need a good firewall and a good anti-virus for complete protection. I hope you have them also!
