The Commonwealth is primarily an organisation in which countries with diverse economic backgrounds have an opportunity for close and equal interaction. The primary activities of the Commonwealth are designed to create an atmosphere of economic co-operation between member nations, as well as the promotion of democracy, human rights, and good governance in those nations.
Political subdivision in the United States is a legal term used to define smaller governmental units which fall under the jurisdiction of the State government. Since each State determines what units of government meet its legislative definition, the definition of political subdivision varies from State to State, and does not necessarily occur in every state.

Attributes of state according to Max Weber: Monopoly of legitimate coertion within a given territory. This means that the only thing a state must have in order to become one is the exclusive right to use the force within it´s territory. In this sence, state is closely related with politics.
A common wealth, on the other hand, relates with economy. It implies certain degree of institutionalized economic interdependence

Puerto Rico is a commonwealth, for ex. Its residents are US citizens, we use US currency and receive a limited amount of benefits because the island is not a full state.
Also in Congress, our representative speaks and can convince others to vote on his behalf, but he can't vote because the island is not a state. A state of the Union has all full benefits, assistance, grants, and pertinent aid.
So basically it is a partial, political relationship

if you mean "commonwealth" as in "Commonwealth of Massachusetts" then there's none - it's just another way to say "state." US law makes no distinction - it has 50 states, not 46 states and 4 commonwealths.
Outside of the US, "The Commonwealth" is a league of Britain and many of her former colonies.
