I think it works, but it's definitely harder then "detecting" gay men because women's sexuality is more fluid than men's. Sometimes, you can tell by the type of look they give you when you pass... I'm gay and I definitely look at women differently than do my straight friends...
If you're seriously trying to meet other lesbians, go to places where you could meet some with whom you may have something in common.

I have no idea. The men's clothes or looking butch thing doesn't work; I have lesbians hit on me often (my significant other refers to me as his butch girlfriend), which then causes me to point them towards my sister, who has long hair and is usually in a dress. Lesbians are tricky to figure out.

there's the stereotype ones (women who dress like guys or who look something like a guy) most likely they are lesbians/ bi.
there are those who don't dress like a guy nor do they dress like a woman, somehow you can't figure out what they are trying to dress like. the probability is good that this category is les/bi too.
there are those who look and act like the rest of the women population. this is difficult to tell, so unless you observe them, then you can't tell if they are les/bi.
A good portion of les/bi work in the art/creative/entertainment/sports industry.
