I read an interview with Tarantino years ago that asked this very question. His first answer was (to the interviewer) "What do YOU think was in the briefcase?" At the end of the interview, the interviewer asked the question again and he said "It was intentionally left ambiguous for the viewer to determine."
Over the years however, the general consensus amongst fans is that it is "Marcel Wallace's soul" or "Macel Wallace's umm, mojo"

It was Marcellus Wallace's soul!! His soul was in the brief case that is why it was so important to get it back!! Notice that the first shot of him was a band-aid on the back of his neck. The biblical thought is that the soul comes from the base of your skull, therefore his soul was stolen and he sent them to get it back!

It was full of gold bricks, hence the golden glow.

Because it was gold, as in the precious metal, probably in bar form.
