when you first turn your computer on, it flashes a black screen
usually on that screen it will say Hit DEL or F2 to enter setup.
hit that key
once you are in the BIOS setup, find the USB and make sure it is enabled.
Depending on the age of the computer, you might have USB 1. If your device is USB2, it will plug in, but not work.
You may need to add a USB 2 card to your PC.
Ask at any electronic/computer store.
If you know what you are doing, you can even get them at WalMart.

Check BIOS first to make sure that they are turned on...consult you manual on how to do this.
If BIOS is OK and USB is ON then I would bet that the driver is not installed or has become corrupted for whatever reason.
Right mouse-click My Computer and go to Properties, Hardware, Device Manager and see if the USP port drivers are installed properly...you can find proper drivers at your computer's maufacturer website OR your motherboard manufacturer website. The install is fairly easy and directions are usually provided on how to do this...
Good luck.

Are you working with XP or an older software? XP can usually pick up on the new installations, but anything older than that it doesn't read.
