The first shuttle launch was STS-1, Columbia, launched April 12, 1981 with a 2 man crew, John Young (who previously flew on Apollo 16 and walked on the Moon) and Bob Crippen.

You could say that the first shuttle was Enterprise and the first man in a shuttle was whoever the test pilot was on it's first manned flight.
But, she never made it to space, so you might not consider her a Space Shuttle. Then Columbia, Challenger, Discovery, Atlantis and Endevour.
