Most tile manufacturers consider plywood to be an acceptable subfloor for installation of tile. You need to make sure the plywood provides a good solid floor with little to no flex of the plywood (the tiles won't flex and this could cause you to have cracks along grout joints). You also need to be sure the surface and joints between the plywood pieces are flat and level. If all this is true, you can then just do your layout on ttop of the plywood and spread the thinset to set the tile on as recommended by the tile manufacturer.
