I`ve used the phrase "A sigh of relief."
Can i use the above too?

relief sounds better to me, because sigh of comfort.. hmh comfort is something you feel but not express in sigh.
you can "sigh in comfort" however.

No. A look of comfort, perhaps. You can sigh in comfort but you are trying to explain sigh as a look.

You can use : His dull face relaxed as he gave a sigh of relief.
