There are 2 theories
1. She was pregnant at the time and her smile expressed the sweet secret she was carrying
2. Da Vinci painted her as androgynous. Amon is the Egyptian god of fertility and Isis(ancient pictogram is L'isa) is Egyptian goddess of fertility. Monalisa is an anagarm of Amon L'isa , a union of male and female. The lady carries this befuddling secret and hence her enigmatic smile.
I like the 2nd theory and have read it in a couple of books incldng the Da Vinci Code.

Mona Lisa was amused Da Vinci thought he was so smart as to hide Mona's pregnancy under a shawl. Mona knew Leonardo didn't father the child; he thought he did.

Hey wait a minute...! How could u tell that she was smiling? Even if she was how could u describe it? To tell u, even the people who r behind the investigation of what actually is the reason behind it have not come to a conclusion whether it is a smile or not, but they have described the smile in many categories it may fall into(eg..happy,sad,emotional,eager,angry,etc...).Lets hope we get an quick answer 4rm them....GOODDAY...

OldTrash is right! Hence the supressed cynical look of the smile,.............or not! DaVinci was a smart aleck, and that has NOT been contested, like so many other things about his work.

There are a couple of theories.
1) that she had just had a baby (hence the shawl she was wearing, that only new mothers wore at that time).

She is pregnant in the picture. That is the real reason for her secret smile, it is the life she is carrying, she is happy about it. Her husband commissioned the painting to be done to show her his love and thanks.
