3 air vents in a certain region of my house emit a terrible (musty, moldy) odor every time my AC kicks on. I've looked in my attic to inspect the air handler and duct work. Everything appears fine as far as I can tell (no obvious mold or mildew, no holes with a dead mouse in it, no intake problem, new filter, etc.), and I don't want to rip out all my air ducts without having some kind of idea as to what might be causing it. Any thoughts?

Moisture sounds like the problem......something that may help kill some of the mildew and mold spores would be to turn the system on, go to the intake vent and spray Lysol while it runs.
Let it get a good dose of that distributed through the system and then, try to locate a place where some moisture could be or could have been present and fix that.

It might not be in the vents. It might be that the mold/mildew is in the room and gets kicked up when the AC kicks on.

If you feel certain it is the vents, see about getting a vent cleaning service in to see if they can find and remove the problem.

The same thing happed to us, it lasted for a few weeks, and went away....We never did find out what the problem was. I did spray Lysol in all my vents
Good Luck

Have one of your filters gotten wet or anything? That can cause odors after it dries out.

Have a technician check for freon leaks. Some freons are propane based.
