Slavery was a normal practice among Indian tribes such as the Cherokee and others prior to the arrival of Europeans. Most slaves were taken as spoils of war during that era. Charleston was easily settled in large part because the small tribes in the Lowcountry of what is now South Carolina were afraid of the three major nations/tribes that regularly raided them for more slaves. However the Europeans enslaved them as their reward; braves were shipped north and bred with captive slaves from Africa while the squaws were bred into the local stock of slaves on the new plantations.

Effectively, if you mean BLACK slavery in the TERRITORY NOW BELONGING TO THE US, that started with the plantation colonization that took place in the late 1600s, the idea of importing African blacks was borrowed from the Spanish who had to do it from the 1500s on when the pope decided that Indians could not be enslaved, and they had killed so many of them in the first place that not enough of them remained to work in the plantations and mines.
