You can learn 'On The Job' or go to travel school... I went to Southeastern Academy in Kissimmee (Florida). We were traind in Travel Agency Functions, Airline Ticketing, Hotel/Motel management, etc. I don't know of any other travel schools, but you might be able to contact Disney (a student in our class was sent there by Disney, had a job waiting, and had her schooling paid in full by Disney) or just search for them on the internet...

You have to have some travel experience, and a bit of experience in sales.

You'll have to learn booking programs such as Sabre, or others. Other than that, I know some travel agents that aren't too bright. I myself never graduated from College.
