I will have my wife with me, so i suspect the red light district is out!!!!

A canal tour, the musueum of gracht, and even the red light district: you don't have to consummate and it IS a touristy spot! The flower market at the end of the main street is worth a visit (especially if you buy your wife a bunch of roses!)
Also great; The beguinage (or Beginhoff - not sure the spelling is correct, check with tourist office called the VVV). Its off the main street and it is an island of peace and quiet.

Walking around in the red light district is still possible. But would you like it?
My advice: A trip with a canal boat (about 1 hour) and a visit to Grand Cafe like De Jaren.
Second possibility: the Rijksmuseum (It is the "rembrandtyear") or the Van Gogh museum. A possibility is a canal bike (near the Anne Frank house, which is also worth a visit), but the weater is maybe too rainy and cold.
All these are around the centre of the town. Just visiting a terrace at the Leidse plein or Rembrandtplein are good, but only when the weater is good.

Visit any of the museums...Rijksmuseum was one of our favorites, but so was the sex museum and Heineken Brewery Experience (plus you get three free beers each!). Check out the Pancake Bakery for lunch or dinner (depending when your layover is) and take in thee Anne Frank Huis (same street).Or head over to the Daam market for some shopping. Then just sit in one of the cafes and enjoy the crowd.

Of course, if you're into it, you can visit one of the many coffeeshops, try the Grasshopper right down the street from Centraal Station or the Bulldog on Oudezijds Voorburgwal. Both offer all kinds of libations, food, & smoke.

Go to the Bulldog cafe for some smoking or the anne frank museum is really good and will only take under two hours to go round, about 7 euros to get in.

You should definitly go for the "free walking tour" of amsterdam... it´s wicked! this guys take u around the whole city (like 3 hours), explannnig all the odd, historical and fuc facts of the city... They gather in front of the main tourist information office (whis is in front of the train station) at 11 am and 3 pm... u will recognize the guys, since the wear red t-shirts!...

When I was there several years ago the fire trucks would circle Dam Square about 6 o'clock every day and water hose the drug heads who were all spralled out on the monument in the center of the square. Need a shower?

Go to the red light district anyway, your wife can comment on their cheap underwear and say that they would do better going to M&S. There are always loads of women walking around there.

Boat trip on the canal is brilliant, and you can take it straight to the door of the Anne Frank museum. It's really interesting...
