I don't know what significant means but I say YES, an inflated tire weighs more than than a flat tire.

An inflated tire is measured by psi (pounds per square inch). If an a tire is inflated to 30 psi, then it contains more air per square inch than normal outside conditions. A flat tire obviously has lower psi (or else it would not be flat). Meaning less air "shoved" into the same space.

Air has weight just like anything else. More air - more weight.

No. Air does have some weight, partly offset by the added volume (air provides a little bouancy). Since air is a about 2 atmospheres gage pressure, it weighs about 3 times aira ambient conditions, but this is still trivial. it will weigh about 3 g/m^3.

There would be some difference but not a lot, the vast majority of weight in a tire is it rim and the tire its self, not how much air is in it.

tires are measured by pressure, not weight. Depending on the amount of rubber used to make the tire itself (thickness, size around...), the weight could vary between different tires, but If you are talking about pressure, usually you will notice a difference visually after a loss of 8-10 cpp (cubic pounds of pressure), but again, that will depend on the size of the tire... relatively speaking (hope this helps)

I remember this experiment from grade 7 science, a deflated basketball and an inflated basketball were place on a balance. The inflated one was heavier than the deflated. So yes, a full tire will be heavier than a flat.
