Is it the seperation of two stands? Am I right?

I think that separation of the strands is the first step.

no, sadly not. :)

the first step would be the uncoiling of the chromosomes.

DNA is usually stored in a coiled up state, doubly coiled to be exact. large proteins called histons serve as spindle and the primary spindles are coiled up yet again on even bigger histons, thus forming the chromosomes that you can see in your microscope.
