The best estimate of the origin of the Universe, from the Wilkinson Microwave Anisotropy Probe, is 13700 million years ago (13.7 Ga). However, at least one subsequent cycle of star birth/death is required for nucleosynthesis of the C, N and O essential to life, and this process may have taken up to several Ga to produce sufficient quantities (Gilmour et al., 1997). This puts the earliest possible emergence of life in the Universe at ~12.7 Ga, although there is large uncertainty in the length of the necessary time period.

It depends on who you ask. As mentioned by an earlier answerer, fossil evidence suggests single celled organisms around 3.5 billion years ago. Interestingly enough, complex multicellular organisms don't appear until around a half a billion years ago. Young earth creationists will tell you that life began 6000 years ago when it was magically created by God.

Nobody knows but we know that most of life's history involved the biochemical evolution of single-celled microorganisms. We find individual fossilized microbes in rocks 3.5 billion years old.
We can identify multicelled fossils only in rocks younger than 1 billion years.
