This is a difficult question, all the islands I have visited are beautiful. Corfu - the drive and views to Palaiokastritsa; Crete - Aghia Nicholaos or the plateau; Rhodes - the view down on Trianda (at sunset especially); Santorini - and all its white houses; to name but a few. So is there really "the most beautiful greek island?"

Santorini would be my 1st chose. The town is way up in the mountain. White a blue buildings. Loads of fantastic locals. And if you are a beer drinker, you will love it. You can either take the stairs up to the city (approx. 500 something stairs though) catch a mule (did that) or take the cable cars.
Then Mykonos is also a fantastic island. Real enjoy every minute there. Very beautiful. The roads are like from a story book.
Small narrow cobble stoned roads. Lots of party life as well.

As has become obvious from the answers so far it is very difficult to chose. There are over 3000 islands in the Greek waters. Each island has something different to offer. It's easier to view them as groups Like the Cyclades, the north and south Sporades the Dodecanese, the Ionian islands Crete, Evia,etc. Depends on what you define as fun. Islands with intensive night life or peace and quiet. Nudism or conservative . Each island has its own subculture from Athens. Each has its own history going back to prehistoric times. Each takes part in Greek mythology. Each has it's own architecture. Most of the islands in the Aegean ar volcanic and therefore the geomorphology tends to be awesome with the caldera in Santorini being the crown Jewel. My advice if you can do it try and visit as many islands within the group and form your own opinion. The island in the Aegean are very closely grouped and you are always within site of (on average) 3-4 islands.

It has to be corfu I've been there 4 times now. Rhodes I went to which is interesting. I can't remember that much but I wouldn't go again because the hotel was infested with bugs, this was 4 star hotel!, and we had to move to 4 different hotels and even then there were ants in one of the beds in the apartment. Corfu is the greenest of islands and if you do go to Corfu visit Paeliokonsticta (I think it is) It is beautiful. I stayed at Rose Gardens in San Stefanos also known as Agios Stefanos which is NW of the Island. There is also a nice beach going from Roda, which I think you should visit! The Views are breath taking and theres lots of places you can go exploring. Corfu town is nice. If you like more developed Towns/cities then you should go to Sidari. Kassopi is really pretty too. I wouldn't go near the south though there's not really much to see. Plus Price Phillip/ The Duke of Edinburgh was born there!

I went Island hopping last Summer and really did think Santorini was gorgeous. It has lots of hotels and houses literally perched on cliff edges due to when the volcano that was there split the island in two. You can also take a walk up the volcano and get a great view of the island. Take a boat trip around and you can see some amazing views and go dipping in the hot springs :-)

Depends what you are after and what appeals to you.
I am half Greek so I can answer, somewhat, unbiasedly...

The best beaches: Antipaxos, Elafonissos in the Peloponese (so wild as well), Chrisi island-south east Crete, Koufonisia (Cyclades), Lefkada (Porto Katsiki, Ag. Nikitas) and Zakynthos (check out Navagio).
For beautiful villages/views/sunsets: Mykonos, Santorini, Amorgos
For nightlife/clubs/bars etc: Ios, Mykonos, Skiathos
For quiet, peaceful time: Milos, Naxos
For lush greenery: Skiathos, Skopelos, Thassos, Crete, Kefalonia
It's so hard to pick, trust me, I've been to over 30 already!

There are so many to chose from it is very difficult, but I would certainly reccomend Kefalonia or Santorini. Both fairly small and not to developed with a real touch of Greece in the small villages. The capital of Kefalonia is Argostoli a good size town with a lovely square on which most of the cafes, bars and restuarants sit.

We have so many islands in Greece, that is becomes merely a matter of taste...
There are dry, arid, naked islands with rocks and beaches only in the middle of crystal clear blue seas, there are islands with pictoresque views of the sunset over steep cliffs, there are islands covered in ancient ruins, speaking of ancient times, there are islands covered in green, full of streams, little rivers, waterfalls and steep waters, there are islands with beautiful small villages and very hospitable people...
You just chose what you want and come to Greece to find it!

p.s. I personally love Samothraki, Santorini and Corfu (not when flooded with tourists).

According to my taste the most nice islands
Serifos, sifnos Milos, Santorini, Lesvos, Chios, Ios ,Folegandros
