That honor goes to the 1899 Cleveland Spiders; that team finished the season with 134 losses against only 20 wins. The 1962 New York Mets would lose 120 games and the 2003 Detroit Tigers would lose 119 games.

To answer your question literally, the Chicago Cubs since 1901 have lost 8,148 games. The Baseball Reference link below has their breakdown to 2005, and I added 2006--96 more losses.
Actually, the Cubs began in 1876, but I couldn't find records before 1901. Their loss total should therefore be a couple thousand more.

In a season, it's the 1899 Cleveland Spiders, who went 20-134 thanks to Frank Robison, their owner, pillaging their team. He had just bought the St. Louis Browns at auction and thought that his stars would draw a better crowd there:

All-time, it's the Phillies, with almost 10,000 regular season losses after 2006.
