I had received an automated e-mail response from one of the jobs, and but have not or will have never received any response from most of the jobs that I had applied for in the beginning. It's been 3 weeks now since I lost my job, so it'll be a month this Sunday. I did receive a response from the recent job I applied for online, and I'll go to hiring site to take the assessment this coming Wednesday.

What would you suggest to increase responses from prospective employers?

My guess: you are applying for jobs where there are lots of other qualified candidates. You need to make yourself stand out. How does your resume look? Like everyone else's? Do you have any special skills that could make you more marketable? (for example, speaking a second language) Have you contacted any of the companies directly, or just sent in your applications? A follow-up phone call can put you in the mind of the recruiter/employer/hirer and increase your chances.

Job market's tight. I just got off of a job hunt, and I'm sorry about what you have to go through. It sucks @ss. Anyway, to increase responses, first of all, make sure your resume includes the same kind of words as the job ad does. Lots of companies use electronic scanners nowadays, so they might be specifically looking for "Microsoft Word and Excel" as opposed to "excellent computer skills", or vice-versa. Also, send a note or call several days later, reviewing what you said in your cover letter and asking if they have any questions. I knew a manager once who claimed she never hired anyone who didn't call after dropping off a resume. Finally, work on your cover letter. Cite examples from previous work experience about how much you rock, and try to sound enthusiastic. Good luck!

More jobs you apply for, more responses you get. Apply for more.

Make sure you have a Resume that is on one page so that they don't have a great deal of reading to do.
