Every disease has it's own vector or what carries it. Bacteria, virus etc. These can be tested for or the antibodies a person infected with them can be tested for.

Blood sample is taken through venipuncture and the blood is centrifuged to obtain the serum. Then ELISA is performed with a small sample of the serum.
