Most Emulators have programmable inputs for them. If you are using NESten you click on inputs on the top bar then click player one. A box will pop up and you click on each input and when it ask for you to press a button then you press the button you wish to be that action.

Learn how to code in C++ and java, download the source code for the emulator, and re-write the code in order to make the USB work with the emulator.
