There are a number of ways:
Run
Pass
Intercept
Punt return
Kickoff return
Penalty
Recover fumble from opposing team
Recover fumble from own team that travels beyond 1st down marker.
Be the recipient of a free kick (which happens after a safety).
There may be a couple more, but those are the ways I can think of off of the top of my head.

there are only 3 ways to get a first down
1 run and gain the necessary yardage
2 throw and catch the ball by the same team and gain the necessary yardage
3 by penalty, some will give an automatic first down some will give enough yards to get the first down

uh...what exactly do you mean?
You can run..pass...get it in 1 play, or 4.
