After Ash beat the Kanto or Jhoto league Misty became the leader of the water gym and Ash went to the Hoenn Region to get more Gym Badges and to ultimately beat the Elite Four. Brock i think became a the profession he said he wanted to become, i think it was a Pokemon breeder or something like that.

Misty's character rarely shows up in the main "Pokemon" series, because she became a Gym Leader. However, if you're watching the dub of pokemon in the USA, you can sometimes see Misty in the "Pokemon Chronicles" series on Cartoon Network. The spin-off show features stories centered around the "minor" characters of the Pokemon show, including Misty.

Misty and Brock decicided to return to thier gyms after a little traveling with Ash and Pikachu eventually you've got have a vacation or you might wind up Gay.

Misty went back to Cerulean City, and become the official gym leader there.
Brock still tags along with Ash, he just has a different outfit.
