In cell biology, the nucleolus (plural nucleoli) is a "suborganelle" of the cell nucleus, which is an organelle. A main function of the nucleolus is the production and assembly of ribosome components. The nucleolus is roughly spherical, and is surrounded by a layer of condensed chromatin. No membrane separates the nucleolus from the nucleoplasm.

Mitosis is a type of cell division. There are other types of cell division, such as meiosis, which is how eggs and sperm develop. Mitosis is the process by which most of the other cells in your body develop. For example, the outermost layer of your skin is made up of dead skin cells. These dead cells are rubbing off the surface of your skin all the time and being replaced by new cells from deeper in your skin. These new cells are formed by mitosis, or one cell splitting into two cells.

If you mean a nucleus as in biological cells, then the nucleus is the "brain" or control center of the cell, it contains DNA, and is responsible for cell-division or self-replication. Obviously, there's more to it than this but in general, that's the nucleus.
