What do you want her to do, pee in her seat? Tell the bus driver to pull over immediately. Also, try going before you get on board.

You mean should they stop the bus? It would be a kind gesture and good business to stop the bus and wait for her to finish and come back. However, most people aren't that kind or patient so it might be too high of an expectation. If the toilet on the bus is broken they need to display a sign on the outside so people would know that they will not be able to use the facilities during their ride.

No, the bus should stop and let her down to pee. Especially if the bus is equipped with a toilet, which means she chose a bus with toilet and it is the bus'es responsibility that it doesn't work.

It depends what kind of bus and the situation. If there is somewhere for the driver to stop, it would be nice, but they can't stop for everyone everytime they ask... you would never get anywhere.
