Honestly and as soon as possible.. Just let him know that while you care about him, you and he are in different places in your lives or drifting apart or you are just not in love with him anymore.. Just try to make it as painless as possible. Start it off by saying - we need to talk, this is really hard for me, I don't want to hurt you and then tell him how you feel - the reason why - it is ok.. He would rather you end it then keep going on in the relationship not happy I am sure.

The best thing is to be straight with him. It's not going to be easy, it never is for a caring person. Don't use the "we can still be friends" line. It just seems patronizing. To someone with romantic interest in you being told you can be friends is like saying "your a nice guy" it just rings hollow. Be reassuring if he asks. Make it clear that it's you, nothing he has or hasn't done, it's just not right for you and you need to move on.

the best thing to do is to just sit him down and tell him face to face there is no sense in holding on to something that isnt there you will just make yourself miserable so just let him know that your feelings have changed and that you dont feel the same way anymore.
