if youre looking for food, head on down to lau pa sat. they have everything you would want to try in one convinient location. it's in town and popular so it's not really difficult to locate at all.
not far from lau pa sat is the merlion at the mouth of singapore river. you can drop by there and snap a few shots as well. there are also boat rides there that take you from boat quay to clarke quay, two popular places teeming with night life. just ask anyone how to get to boat quay and they'll be happy to assit. it's really close by.
for shopping you can make your way to orchard road. to be honest though i find this place a little dull. but then again i'm never one for shopping.
since you only have a day, i recommend culture soaking:
for sightseeing, you can head on down to little indian to catch the lights that are put up for the upcoming deepavali celebrations. you can also try some indian cuisine there. you can also visit the one fo the many temples there.
make your way down to geylang serai in the evenings for more food (malay cuisine this time) and shopping.

There's so much to see and do in Singapore, a single day is not enough.
Shopping is everywhere and eating at their 'food courts' (found in most shopping centres) is quite an experience, great selection of cuisines of the many ethic groups living there.
Best bet for your single day is to hop on one of the city tours - see/experience many facets of Singapore in the shortest time possible. Check that out at the Airport/Arrival, there's something for stayover passengers.

You can hardly do anything fruitful within a day in Singapore. Shopping alone will take you more than that - depending on how you shop of course.
Food sampling will also be time consuming. Then again the sight seeing.
For a quick one, maybe just visit Orchard Road to do some quick shopping and a little food.

..Pretty for good food, it's an exotic experience of being able to try Asian food, from Pakistani, Northern and Southern Indian food, Vietnamese to fusion food all at one go, only in Singapore. Try Little India for typical Indian, Suntec city offer plenty food and shopping experience. For electronic such as computer, try Sim Lim (please bargain), Mhd Mustapha supermarkets is a good place for everythings, including memoirs. Food must try local specialities like Chicken rice, laksa.... ask any taxi driver, they will know. They can help you to the inexpensive shopping and food areas to those that are premium but also offer good food and services. Singaporeans are all friendly lots. Very safe, just need to open your mouth and ask.... even roadsider will help you...enjoy your trip.

Only one day in Singapore?
The airport offers a cheap (maybe still free) City Tour for transit passengers. Check it out at the transit desk before you go through the customs!
The best food is in Little India Area, lots of nice squares where you can pick your choice and eat outdoor. And plenty of nice restaurants too. You can take a public bus from the airport to Bencoolen Street, if you wish to save money for a taxi.
Little India is great fun for wild souvenir shopping and exotic snacks.
Orchard road is good for more serious shopping, if you are looking for camera, laptop, etc.

One day? Dont expect to see much in 1 day not here. You can shop at the airport. Food there should meet your requirements.
Singapore is not a one day city. There is much more in Singapore to experience. I suggest you take a good rest in the hotel, can come back when u really feel like enjoying yourself.
