Best sci-fi book is "The Stars my Destination" by Alfred Bester IMHO. It's set in a few centuries in the future, when people have developed the ability to teleport using only the power of the mind, a process known as 'jaunting'.
What makes the book great is that it's actually kind of believable, despite the absurd (even for sci-fi) premise.
Although written in 1956, this is way ahead of its time and is credited as the original 'cyberpunk' novel - the plot and atmosphere are very similar to William Gibson's 'Neuromancer'.
Oh, and it's sometimes published under the alternative title "Tyger, Tyger".
Essential reading if you're into sci-fi.

The Crystal Singer series by Anne McCaffery
Any of the Dragons of Pern books by Anne McCaffery
But the best of her books have to be the ones in the series which includes The ship who sang, these are totally awesome and are about really disabled kids who's brains are fused with either a ship or a whole city so they get to live a different sort of life amongst the stars or on a colonised planet accompanied by a regular human in their adventures. Anne Mc Caffery rules.

I'll chime in for Herbert's Dune as well. The writing is great, comparable to literary fiction, and the depth of the universe he creates is rarely matched by sci-fi writers. He's awesome. I think the underrated God Emperor of Dune is great as well, though the series loses its steam a bit in the last two books.
The books his son and another guy have written to cash in on the Dune legacy are badly written and kind of boring; don't bother with them.
