What is the proper response for receiving flowers? Should I send a thank you card or just a quick note?

How you respond would depend on why you received them.
However I don't think you can ever go wrong with a thank you card.

Make it both. Rather than write a lengthy thank you, just say "Thanks very much for the flowers--they're beautiful!"

Whether or not you send a "real" thank you card through the mail, make a phone call, or send an email depends on your relationship with the sender. In my workplace, it is appropriate to say thank you in person, if there was one giver, or send a group email if the flowers came from a group.
