Actions speak louder than words.
Examine what you do to provide a successful work environment. Then look at what you do to put up obstacles in her path.
Talk is cheap. And secretaries have been on the short end of the payscale forever. Seriously consider a raise, or at least a bonus especially if there has been long projects, etc... Believe me, when I tell you, whatever you're paying her isn't enough.
I had a boss once who thanked me constantly. It didn't help as he was inconsiderate and a poor time manager.

In what way?
If she feels that she is not significant in her role in the company, then some encouragement and positive words can go a long way. Something to the effect of, "I really appreciate your effort on that last project, you are a great help." Note the use of "are" as opposed to "were".
Now if you are speaking of trying to date her, BE EXTREMELY CAREFUL. The best rule of thumb is not to date someone that you work with. Unfair as it has become, a woman needs only to mention "sexual harassmant" and the guy becomes guilty until proven innocent. Her word against yours, and guess which side they take, if only to cover their own back sides from a potential lawsuit and bad publicity.
However, if you want to ask her out, a company party or other setting where there are several other members of the company is one way to chat with her in a more relaxed environment. You'll have to play it cool and see if she seems interested in you.

I take it you mean in a business sense
Next employment anniversary, secretary's day or what ever reasonable occasion
give her a day treatment to a spa or a gift certificate to a nice restaurant, or better still at a real fancy restraint arrange to have the meal for her and her other paid (by YOU) make sure she realizes you foot the bill even if the company covers it later - for instance the maitre'd could say yes Ms./Mr.>>>> , Mr/Ms. >>> has instructed us to take special care of you this evening.(tip the guy extra to make sure of this point.
