Check the oil with the engine off.
Nobody can tell you how to check trans without knowing what kind of car you have.

You always check engine oil when car is cold and hasnt been driven for at least 3 hours. Transmission oil is always check when the car is thoroughly warmed up. A good 10 miles drive in stop and go traffic should do the trick

Check the oil with the car off. Wait about 3 mins so the oil has time to run back into the pan. Pull out the dipstick wipe it clean,reinsert it all the way in,pull it out and check the level.
The transmission fluid is checked with the car running. The instructions for weather it should be in park or neutral are written on the dipstick. Pull the stick out wipe it. Read to see if you need it in park or neutral. Reinsert dipstick all the way pull out and see where the level is.

First have the vehicle on a level surface when checking either.
Check the oil when the engine is off. Warm or Cold.
Check transmission fluid with the engine running, transmission in neutral, and the parking brake on.

Check your engine oil when the engine is cold. Check the auto transmission fluid after the engine warms up so that the proper level is what you will have on the pan and keep engine running. You may put the gear on D first and back to Park before checking and make sure the dip stick is clean before doing so. But first and most importantly put the parking brake on.

wow tony got it rite as some hondas check the trans fluid with engine off
oil engine off all the time most cars engine running for trans
be sure to find out if your car uses a special oil in the trans dodge and chev do that noy
best bet check the owners manuel thats what there there for
