Great place for live music and great food, particularly if you enjoy BBQ or Tex Mex.
I'd advise scheduling at least 1-2 nights for 6th Street/Downtown (the warehouse district or the 2nd St districts are a little bit "hipper" than 6th, but more pricey) and try and see a couple of shows as well. The Austin Chronicle has listings and reviews for clubs and restaurants, so that's a good place to start your search. Personally, I like seeing shows at Stubbs (outdoors), Antones, and the Continental Club, but they usually aren't seated shows.
There are TONS of good Tex-Mex restaurants (it's very different from Cal-Mex or interior Mexican food) and several good BBQ restaurants. I'd check the Chronicle for your favorite type of food, and read the reviews. Chuy's is usually a favorite of both locals and out-of-towners, as are Shady Grove (really just "Texan" food, not Tex-Mex), Maudie's, and Trudy's.
If you like the outdoors, try and spend some time at the lake. It's nice, but usually pretty hot. If you're into pretty scenery, I'd check the Texas Parks and Wildlife website for some of the nearby places you can see wildflowers and other nature-y stuff.

Day activities-Zilker Park or Lake Travis-Lady Bird Johnson Wildlfower Research Center, State History Museum or the DaVinci Exhibit at the Art Museum across the street from the history museum. Tour the capitol and governor's mansion for some state/local flavor.
Evening-Definitely 6th street, Esther's Follies for comedy, but there is almost ANY food or venue of entertainment you could think up. Some other fine restaurants also on Congress either side of 6th.
Mind blowing experience-the new Whole Foods Market at 6th and Lamar. But I like Central Market better-less cluttered, confusing, but amazing choices.
