There are so many. I love my Frank Capra movies though, so probably either It's a Wonderful Life or Mr Smith Goes to Washington.
I like Night of the Hunter too, but it's a very different movie so I couldn't really say it was better or worse than those two.

It has to be "It's A Wonderful Life" with James Stewart & Donna Reed. I still cry at the ending, no matter how many times I've watched it! Absolutely brilliant!!
