Conformation os the shape of the active site of the enzyme, aka the 3D conformation.
Denaturation is a irreversible process and the enzyme cannot catalyse reactions (cannot work anymore) as the shape of the active site is changed by heat/high and low pH as bonds are broken

Enzymes denature do to use and heat, after a period of time they conform or return to there useful state.
