Yes, as has been noted, there was at least one mouse in the story, but that is beside the point. The title is taken from a line in a poem by Robert Burns, not Robert Frost, "the best laid schemes o' mice an' men gang aft a-gley." Or, in other words, "the best laid plans of mice and men often go wrong." Thus the title refers to the plans the guys had to save up their money and buy a little place of their own, where Lenny could tend the rabbits, etc. Something unforseen happened that completely derailed their "best laid plans."
Oh, all right, I suppose the mouse that Lenny killed might have had "plans," too, but they certainly went "a-gley."
