Theoretically, you could do that by putting an object with equal gravitational pull above you. That means either another planet or a black hole balanced upstairs from your gravity room. In practice, that gets a bit hard to engineer...

As far as is known, everything has positive gravity: antimatter, Dark Matter, supersymmetric particles, even Dark Energy (although it is pushing the Universe apart, but that's another story). And nothing "shields" against gravity. So there is nothing you can put on the floor to cancel out the Earth's gravity. You could cancel out the gravitational force with electromagnetic fields, for example by giving yourself a big electric charge and suspending yourself in a giant capacitor. This wouldn't feel exactly like zero gravity, because the charge would all go to your skin and your skin would be pulled up while the inside of your body sagged down.

The only hope for real anti-gravity is to put an enormous, dense mass in the ceiling. Even so, this would cancel gravity only at one height in the room---if you floated upwards from that height, you would stick to the ceiling, and if you floated downwards you would stick to the floor (like you're doing now).

NASA simulates zero-g by floating the astronauts in big water tanks.

I can tell u a simple way Just wear a metal jacket and apply an electric field in the anti - gravity direction such that ur weight equals the attraction of the electric field resulting in weightlessness.Once this is done u'll feel lighter than ever in this condition ur weight is zero.Thus apparently gravity is zero.If u want to move against gravity just increase the field value so that it would pull u away from earth surface.

You can't. As far as we know, there is no such thing as anti-gravity. You can achieve temporary weightlessness by being in a container of any kind that is droping freely through the air. What happens is that the both you and the container are being pulled down, so you do not feel any force pushing you up when you touch it's bottom thus the illusion of 0 gravity is created.

You cannot create an "anti gravity room" anywhere. That is, we have yet to find any material that blocks out gravity (gravitons). And the theories we work with doesn't exactly predict any such material.

You will however experience 0 g during a free fall (if wind isn't taken into consideration)

The force of gravity is based on two things, the masses of the objects involved, and the distance between them. Gravity is a completely seperate force from electromagnatism, so as far as we know, there is no way to use EM fields to alter the force of gravity. The only way to do that is to change the distance between objects, or change their mass.
