It takes years and years of practice. Some people have a knack for understanding others, some don't. If it doesn't come naturally, you're going to need to study body language, psychology and other topics related to human nature. Only then can you begin to understand people -- and even then some people will never be understandable.

Understanding people does take a lot of experience but maybe u can have some idea if u :
1. try to categorise and grade the people you know on their personality traits viz. dedication, sincereity, truthfulness, positive attitude, leadership qualities, problem solving ability etc.
2. try to create a prototype qustions that you can ask these people which would help you study and analyse the responses
3. using the information that you have gained form the above exercise experiment with a sample
also when ever you meet a new person try to find out if you can judge them and then verify if possible whether you assumptiuons are correct.

Some people have the natural ability to read others, it comes very valuable when you end up managing people.
I learned the hard way: I enlisted in the US Army. My role model was a complete **** but at the same time was the most fair person I have ever met wearing a uniform. He was also rabidly defensive when it came to people messing with his team. People that worked in other teams thought it sucked to work for him because he was such a ****, but those of us that had the honor to work for him knew that as long as we followed orders and did our jobs he would defend us better than our own parents.
From him I learned to protect my people. There is a lot of political maneuvering going on in any big company, so as a manager one of your jobs is to protect your employees from all that crap so they can do their jobs in peace. If somebody from outside my team had a problem with my guys, they had to come thru THEIR manager, then that manager had to talk to me and I took the proper action.
Take care of your people and they will do whatever you want them to. The rest will come on its own as you get used to manage.

Be a good listener first digest what they are saying ask them what would they do in such a situation weigh the pros and cons in the interest of the organisation then decide what to do .Next pick up some good skills like body language, you can also read a person through his hand writing,etc.

To understand people, put yourself in their shoes. Your perspective may change and I am sure you are smart enough to find the words then. Try simply ask politely "Interesting you say that but can you tell me why"? You will be surprised how a conversation may begin. This also reminds me of a joke....
They say to understand a person, try to wear his shoes and walk a mile. In case the guy gets mad at you, at least you are a mile away and he has no shoes. Sorry, just can't pass that one up.

Good Question. It is difficult to say human nature.One morething you'n not mentioned ,where? In interview:- approach, fearness,innocence,subject knowledge, reply movement can be justify.Personal contact time:- their urgency, guiltyness,obey , respect etc.,In your boss:- It is difficult to say, how to be handled with him, because moment of mood cann't be determine. You are telling management catagory, separate chapter are there,you may reffer phsicology of human being, their body language etc.,
