Actually the answer is yes, the distance between two points can be different when using the same ruler. You actually see an example every day and it's a household item. However in mathematics if the points are on a three dimensional curve the points can have any number of different distances between them. Cut a rubber ball in half, get a ruler made out of sting and put two points anywhere on the circle where you sliced the ball in two. Measure where you made the cut and the surface is flat on the bottom of the ball you will get one answer. As you move the sting ruler up towards the top of the ball the distance will increase incrementally until you get to the top and then start to decrease again as you move down again over the top of the ball.

Yes, if the points are on a curved surface. You could measure the distance between, say, London and New York (with a really big ruler) by drawing a line around the globe, but it would be much shorter to measure the distance by cutting a line straight through the planet.

Yes.
You measure once in the direction of movement and the second time in the opposite direction. Of course you have to travel close to the speed of light.

the only way that the distance between two fixed points can be different is if there is a bend in space..
