I have a long list of possible grant sources. Is it wise to send out a bunch of Letter of Inquiries to multiple companies at once or should I stagger the submissions?

The sooner you send them out, the sooner you'll get your answers. There's no reason to withhold sending your inquiries. You'll find that there may be different submission dates or deadlines for the actual proposals.
