Diagnostics test showed that a lamba sensor needed replacing,i did that and it ran perfect for about 300 miles,then it started to show the yellow engine warning light and misfire,can anyone help please.

spark plugs and or coils are failing. Take it to a shop that has a Mercedes Benz scan tool to diagnose it properly.
After Market scan tools will not show you the proper diagnostic procedure to repair your 600 the first time correctly.
You spent the money on the car when you purchased it, now spend you money wisely and have it fixed at a shop that fixes MBenz's on a regular basis.

Did you also check the spark plugs and spark plug cables? They might be the cause.
