Actually, unlike a previous answer, the whole of Congress is the House of Representatives AND the Senate.
Each State has different laws/rules to determine how to replace them. Some allow the Governor to name replacements, others call for elections. I actually believe the US Constitution give the States the right to decide replacements. A replacement is only good for the remainder of the elected term of whoever you were replacing. So if you were chosen to replace a Senator whose term runs out in two years, that's how long you would serve. Unless you got re-elected.

well, actually, the President "runs" the country. But he, is just "one third" of the government. The judiciary makes up a third, and the last third is Congress.
So, in the unlikely event that something horrific happened, and all the members of Congress (both the House and the Senate) were killed. I would imagine that a period of national mourning would take place. Then a nationwide special election would take place - to fill the spots of the deceased members of Congress.
I would imagine that there would be some sort of agreements made so that the other two branches of government (the executive and the judicial) didn't take the opportunity to "gang up" on the now weaker third branch.
But it's all speculation.
